Research areas:
The overall aim of the research carried out in our lab is to develop targeting drugs for cancer therapy and identify novel biomarkers for cancer diagnosis.
Research Area 1---Identify antibodies or peptide ligands against tyrosine kinase receptors.
Drugs targeting against tyrosine kinase receptors become important agents for cancer therapy. One of our interests is to identify some antibodies or peptide ligands against receptor over-expressed in cancer or mutant receptor expressed only in cancer. We are now making some monoclonal antibodies against EGFRvIII (a mutant form of EGFR), IGF-1R and Tie-2. Using phage display, we have identified novel peptide ligands for EGFR and Tie 2. Recently, we have developed a novel phage display method (Shi B, BioTechniques, 2007). Using this method, we have identified some scFv antibodies against EGFR and EGFRvIII. Currently, we are 1) to genetic engineering the antibodies against EGFRvIII; 2) to determine the anti-tumor effect of the antibodies; 3) to make immunotoxin against EGFR and EGFRvIII; 4) to radiolabel these antibodies for caner diagnosis and therapy.
Research Area 2---Develop receptor-targeting nanoparticle and liposome
Receptor-targeting nanoparticle and liposome are promising vectors for delivering drug into cancer cells. On the basis of those antibodies and peptides identified by us, we are now trying to make targeting nanoparticle and liposome. Nanoparticles are made by displaying peptide ligands on viral capsid particles derived from HBsAg. Immunoliposome are also being made by conjugate antibody fragments to liposome. The immunoliposome was used to carry chemical drugs and therapeutic genes for killing cancer cells.
Research Area 3---Develop Cancer Vaccine
Cancer vaccine has become a promising way to kill cancer. We are now using DC vaccine and peptide vaccine to kill some kind of solid tumor.
Research Area 4---Develop autoantibody profiling for the cancer diagnosis and therapy.
Cancer early diagnosis is critical for the prognosis. However, the tumor associated antigen was minimal in early stage cancer patient. The method available now is not sensitive enough to detect these minimal tumor associated antigen. Since, minimal antigen can induce lots of antibody, we are now trying to develop autoantibody profiling in the cancer patient.
